Abstract Background During acute rejection of organ or tissue allografts T cells and macrophages are dominant infiltrating cells. CD4-positive T cells are important for the induction of allograft rejection and macrophages are important effector cells mediating cytotoxicity via production of nitric oxide (NO) by the inducible NO-synthase (iNOS). In the present study we analysed whether the destruction of primarily nonvascularised parathyroid allografts is also mediated by iNOS-positive macrophages. An intravital fluorescence microscopic method for measurement of changes in macromolecular permeability has been established in the mesenterial microcirculation of the rat. After exteriorization of the fat-free distal part of the ileal mesentery, a 1-hr period of stabilization was followed by the injection of FITC-labeled macromolecules. Five minutes later, histamine, leukotriene B4, or leukotriene C4 was topically applied to the tissue by means of a micromanipulator. Areas of 1 mm2 were videotaped with a SIT camera. Whole pituitaries or adenohypophyses alone of adult female Wistar/ Furth rats were dissociated into single cells by means of two different enzymic disintegration methods. The single-cell suspension was then seeded out and cultured for up to 8 months in tissue culture dishes with untreated and polylysine coated surfaces. The cells were cultured in different sera (horse serum, newborn-calf serum, fetal-calf serum, mixtures of horse and newborn-calf serum, and isogenic rat serum) and also in a serum-free, hormone-supplemented medium.
