Analysis of parathyroid graft rejection suggests alloantigen-specific production of nitric oxide by iNOS-positive intragraft macrophages

2009

Abstract Background During acute rejection of organ or tissue allografts T cells and macrophages are dominant infiltrating cells. CD4-positive T cells are important for the induction of allograft rejection and macrophages are important effector cells mediating cytotoxicity via production of nitric oxide (NO) by the inducible NO-synthase (iNOS). In the present study we analysed whether the destruction of primarily nonvascularised parathyroid allografts is also mediated by iNOS-positive macrophages. Methods Hypocalcaemic Lewis rats received parathyroid isografts (from Lewis donors) and allografts (from Wistar Furth donors), respectively, under the kidney capsule. Levels of serum calcium above…

Xid-defective male (CBA/N x C57BL/6)F1 accessory cells present bovine insulin to long-term cultured F1-restricted T-cells

1982

The reactivity of H-2b-restricted murine T cells towards bovine insulin was reported to depend on the expression of Ia. W39, a private specificity of I-Ab, on antigen-presenting cells. Cells of male (CBA/N X B6)F1 mice carrying the mutation xid on the X chromosome lack IA. W39 on the cell surface. These cells are unable to present bovine insulin to primed T cells derived from female (CBA/N X B6)F1 mice. We show here that spleen cells of male (CBA/N X B6)F1 hybrids served perfectly as accessory cells for the insulin-dependent induction of a proliferative response of long-term cultured T cells with (B10 X B10.BR)F1 genotype, restricted to recognizing insulin in the context of F1-unique I-A de…

Mediator-Induced Changes in Macromolecular Permeability in the Rat Mesenteric Microcirculation

1995

An intravital fluorescence microscopic method for measurement of changes in macromolecular permeability has been established in the mesenterial microcirculation of the rat. After exteriorization of the fat-free distal part of the ileal mesentery, a 1-hr period of stabilization was followed by the injection of FITC-labeled macromolecules. Five minutes later, histamine, leukotriene B4, or leukotriene C4 was topically applied to the tissue by means of a micromanipulator. Areas of 1 mm2 were videotaped with a SIT camera. The fluorescence intensity of these areas was measured by an analogous video image processing system and displayed as gray value histograms. The shift of the frequency of gray …

Contracting striated muscle fibres differentiated from primary rat pituitary cultures.

1982

Whole pituitaries or adenohypophyses alone of adult female Wistar/ Furth rats were dissociated into single cells by means of two different enzymic disintegration methods. The single-cell suspension was then seeded out and cultured for up to 8 months in tissue culture dishes with untreated and polylysine coated surfaces. The cells were cultured in different sera (horse serum, newborn-calf serum, fetal-calf serum, mixtures of horse and newborn-calf serum, and isogenic rat serum) and also in a serum-free, hormone-supplemented medium. When the cells were cultured in medium containing horse serum (15 %) plus fetal-calf serum (3%) on polylysine-treated surfaces, cell fusion and the development of…
